Enarodustat hydrochloride (JTZ‑951 HCl) is an orally‑active small‑molecule HIF‑PH inhibitor. By inhibiting HIF‑prolyl hydroxylase enzymes (PHD1, PHD2, PHD3), it stabilizes hypoxia‑inducible factor‑α (HIF‑α) under normoxic conditions. Stabilized HIF‑α up‑regulates transcription of target genes, stimulating endogenous erythropoietin (EPO) production from kidney and liver; meanwhile it down‑regulates hepcidin, improves intestinal iron absorption and mobilizes stored iron, comprehensively correcting renal anemia. It is developed for once‑daily oral administration. It raises hemoglobin in a physiological EPO concentration range, differing from conventional injectable erythropoiesis‑stimulating agents (ESAs).

Important safety note

Risk of rapid hemoglobin elevation, hypertension and thromboembolic events. Not recommended for patients with active malignant tumors. Contraindicated in pregnancy and patients with known hypersensitivity to the compound.
