Naftopidil dihydrochloride is a subtype-selective α1-adrenoceptor antagonist API for men’s urology, indicated for lower urinary tract symptoms (LUTS) caused by benign prostatic hyperplasia (BPH)PMC. It exhibits preferential affinity for the α1D adrenergic receptor, together with moderate 5-HT1A agonistic activity and mild calcium antagonistic effect, which differentiates it from other α1-blockers such as tamsulosin.

By relaxing smooth muscle of the prostate and bladder neck, it relieves both voiding symptoms (hesitancy, weak stream) and storage symptoms (frequency, nocturia, urgency). It shows less risk of certain class-related adverse effects including ejaculatory dysfunction and intraoperative floppy iris syndrome, making it a well-tolerated option for BPH treatmentPMC.
