Tolebrutinib
Code: SAR442168, PRN2246 Tolebrutinib is an oral, brain‑penetrant, covalent and selective Bruton’s tyrosine kinase (BTK) small‑molecule inhibitor originally discovered by Principia Biopharma and later developed by Sanofi. It irreversibly binds to BTK and suppresses the activation of B cells and microglia within the central nervous system.
BTK mediates key immune signalling in B lymphocytes and myeloid cells. Aberrant activation of these cells drives neuroinflammation, demyelination and axonal damage in multiple sclerosis (MS). Tolebrutinib crosses the blood‑brain barrier to inhibit BTK in both peripheral immune cells and CNS resident microglia.
It has been approved for adult patients with non‑relapsing secondary progressive multiple sclerosis (nrSPMS) to slow disability accumulation. It was also investigated in relapsing forms of multiple sclerosis in Phase 3 clinical trials.
