Mechanism of Action

Tezacaftor is a CFTR corrector. It improves protein folding, cellular processing and intracellular trafficking of mutant CFTR protein (including F508del mutation), increasing the quantity of mature CFTR protein transported to epithelial cell surfaceDailyMed. It is generally used in combination with ivacaftor (CFTR potentiator) or elexacaftor+ivacaftor triple‑combination regimen to restore chloride‑channel function, reduce thick mucus accumulation in lungs and improve cystic‑fibrosis clinical symptomsPMC. Compared with lumacaftor, tezacaftor shows less CYP‑mediated drug‑drug interaction and better safety profilePMC.

Indications

Tezacaftor is used in combination therapy for the treatment of cystic fibrosis (CF)DailyMed:

1. In combination with ivacaftor (Symdeko / Symkevi): for cystic‑fibrosis patients aged 6 years and older, who are homozygous for the F508del mutation or carry responsive CFTR gene mutationsDailyMed.
